I know Prototype is still widely popular as the original in condensing the syntax for writing Javascript. Increasingly popular jQuery is has taken condensed syntax to a whole other level. I have recently noticed that the Ext library has grown in popularity and now has it own base library -- or can be used with Prototype or jQuery. Both Prototype and jQuery have user interface libraries, but Ext again takes the UI features to a next level.
I have kept it to these three, but if there are other libraries that many people use let me know. I will be glad to add them so you can then vote for your favorite.

Mootools, not sure why, but it is my weapon of choice.. always.

Don't you mean javascript? 
I used to be a prototype / scriptaculous guy, but Kieran Huggans turned me on to jQuery and I haven't looked back. Occasionally I use MooTools though because they have some REALLY sexy effects that I just can't find in jQuery.
